Calpainopathy is characterized by symmetric and progressive weakness of proximal limb-girdle muscles. The age at onset of muscle weakness ranges from two to 40 years. The phenotype shows intra- and interfamilial variability ranging from severe to mild.

How rare is LGMD2A?

LGMD2A is likely the most common autosomal recessive LGMD, accounting for up to 30% of all recessive cases.

What causes LGMD?

LGMD is caused by mutations of genes that result in abnormal function of proteins in muscles, thereby causing progressive muscle weakness. The inheritance pattern of LGMD varies and different genetic defects may result in the same clinical symptoms in different patients.

How common is LGMD?

The prevalence of LGMD is unknown, but estimates range from one in 14,500 to one in 123,000. The age of onset can vary greatly even among individuals of the same family. The relative frequencies of the different types of LGMD vary from population to population, but worldwide LGMD2G, 2H and 2J are extremely rare.

Milder forms present with symptoms other than weakness, such as muscle aches, cramps, or exercise intolerance, and people in this group can retain ambulation beyond age 60.
