Can a Lexus front door lock and unlock?
Only the passenger’s side rear door will lock and unlock. It’s annoying, but i’m not sure if this is a programming thing or if this require door actuators all the way around.

You should find it at the top of the door panel. From the interior door handle, you should be able to trace the lock rod from the lock, to the back of the door, closer to the latch where you will find the actuator at the other side of the lock rod. Disconnect the electrical connector.

Is there a key to unlock the front door?
The key will unlock the front passenger door. It’s just the remote that will not open or lock the door. I am having the exact same problem with my driver side door. It is a 2003 RX300. If I find out how to fix it, I’ll post back.
