What is MethoCult?
MethoCult™ H4434 Classic (MethoCult™ GF H4434) is a complete methylcellulose-based medium for the growth and enumeration of hematopoietic progenitor cells in colony-forming unit (CFU) assays of human bone marrow, mobilized peripheral blood, peripheral blood, and cord blood samples.
What is CFC blood test?
The colony forming cell (CFC) assay, also referred to as the methylcellulose assay, is an in vitro assay used in the study of hematopoietic stem cells. The assay is based on the ability of hematopoietic progenitors to proliferate and differentiate into colonies in a semi-solid media in response to cytokine stimulation.
How do you defrost MethoCult?
Thaw MethoCult® Express overnight under refrigeration (2 – 8°C) or at room temperature (15 – 25°C). Do not thaw methylcellulose-based medium at 37°C. 2. Once thawed, shake the bottle vigorously for 1 – 2 minutes, or until the medium becomes opaque with bubbles.
What is the difference between CFU and BFU?
Whereas BFU-E cells are CD34-positive, CD36-negative, and CD71 low, CFU-E cells are CD34-negative and both CD36 and CD71 are positive. Thus, human BFU-E cells are CD45+GPA−IL-3R−CD34+CD36−CD71low, whereas CFU-E cells are CD45+GPA−IL-3R−CD34−CD36+CD71high.
