What is Masson trichrome stain used for?
PURPOSE: Used to differentiate between collagen and smooth muscle in tumors, and the increase of collagen in diseases such as cirrhosis. Routine stain for liver and kidney biopsies. PRINCIPLE: As the name implies, three dyes are employed selectively staining muscle, collagen fibers, fibrin, and erythrocytes.
What staining is best for lipid detection?
The oil red O (ORO) stain can identify neutral lipids and fatty acids in smears and tissues. Fresh smears or cryostat sections of tissue are necessary because fixatives containing alcohols, or routine tissue processing with clearing, will remove lipids. The ORO is a rapid and simple stain.
Why is Trichome the best stain?
Staining differentiates tissues by tinting them in contrasting colours. It increases the contrast of microscopic features in cells and tissues, which makes them easier to see when viewed through a microscope. The word trichrome means “three colours”.
How do you make Masson trichrome?
Procedure:
- Deparaffinize and rehydrate through 100% alcohol, 95% alcohol 70% alcohol.
- Wash in distilled water.
- Stain in Weigert’s iron hematoxylin working solution for 10 minutes.
- Rinse in running warm tap water for 10 minutes.
- Wash in distilled water.
- Stain in Biebrich scarlet-acid fuchsin solution for 10-15 minutes.
Why are histological sections stained?
Staining is used to highlight important features of the tissue as well as to enhance the tissue contrast. Hematoxylin is a basic dye that is commonly used in this process and stains the nuclei giving it a bluish color while eosin (another stain dye used in histology) stains the cell’s nucleus giving it a pinkish stain.
Which fixative is used for lipids?
Certain special fixatives, e.g, clichromates and chromic acid, are used for fixing or immobilising lipids; those fixatives ire used for fixation of tissues containing a high concentration of lipids, particularly of nervous tissue.
Which staining method is used for demonstrating intracellular lipids?
Lipid staining is a useful technique that is used for demonstrating intracellular lipids in various tissue sections.
What type of collagen does trichrome stain?
type I collagen
In histology, the trichrome stain is used to confirm the presence of type I collagen and is commonly used to interpret liver biopsies.
What Colour does Masson trichrome stain?
blue
Masson’s trichrome. Nuclei and other basophilic (basic-liking) structures are stained blue, cytoplasm, muscle, erythrocytes and keratin are stained bright-red. Collagen is stained green or blue, depending on which variant of the technique is used.
