Either the power tubes or the rectifier tube. Pull the power tubes, replace the fuse and turn the amp on. If it blows again it is the rectifier tube. ^Do you mean to just pull the power tube and leave them out and turn the amp totally on and see if it blows? or switch power tubes and try again?
